New properties of stem cells via simulated microgravity

ScienceDaily (June 29, 2012) ? A recent study led by Andrew Puca, Ph.D. under the supervision and direction of Antonio Giordano, M.D., Ph.D. set out to illustrate novel mechanical transduction properties of Hematopoietic Stem Cells in relation to defining the expression of humoral factors by facilitating paracrine/autocrine signalling via microgravity.

Dr. Puca, lead author of the study, says "The goal ultimately was modulating intracellular trafficking of tyrosine kinase receptors, cellular proteins directly involved in vasculogenesis, in the context of cell and tissue morphogenesis, ultimately emphasizing the properties of mechanical loading via simulated microgravity and its effects on hematopoietic stem cell (HSPC) growth. These mechanisms are vital to understanding the cooperative relationship between cells and their surrounding microenvironment.

Understanding the expression of these angiogenic triggers will prove to be the cornerstone of bioengineering and the mechanistic machinery that induces tissue regeneration. Our studies indicate that living cells are literally hard-wired so that they can filter the same set of chemical inputs to produce different functional out-puts and this mechanism is largely controlled mechanically," said Dr. Puca, a Postdoctoral Fellow at the Sbarro Institute for Cancer Research and Molecular Medicine located at Temple University in Philadelphia, PA.

"Our data show how cooperation and competition between extrinsic micro environmental chemical signaling (biochemical) and physiological stresses (bio-mechanical) may be the new frontier in delineating processes in stem cell differentiation and improving techniques in tissue engineering as well as provide a new a model for the development of innovative culture techniques for cell transplant based therapies," says Antonio Giordano, M.D., Ph.D., Director of the Sbarro Institute for Cancer Research and Molecular Medicine and Center for Biotechnology at Temple University, and President of the Sbarro Health Research Organization, Inc.

Some South Florida healthcare leaders applaud Supreme Court ruling

Several major South Florida healthcare leaders applauded the Supreme Court?s decision Thursday on the Affordable Care Act because they say it will bring more access to the region?s large number of uninsured and allow for providers to get paid for their care.

But doctors leading the Miami-Dade and Broward medical associations expressed concerns about whether physicians will be reimbursed enough under the new legislation. The leader of Florida Blue saw some positives while warning of increased healthcare costs ahead. Jackson Health System foresees considerable reductions in funding. And individual reactions ranged from calling the ruling a life-saver to decrying the decision as creating rampant socialism.

The stakes are huge for the region?s uninsured -- 31.8 percent of Miami-Dade residents, 24 percent in Broward and 32 percent in Monroe County, according to 2010 Census data. The national average is 16.3 percent. Among adults aged 18 to 64, the potential effect is even more stunning: 57 percent uninsured in Hialeah, 50.4 percent in the City of Miami, 48.5 percent in Deerfield Beach. Even in middle-class Kendall, almost one-third -- 31.2 percent -- of adults aged 18 to 64 now have no coverage.

?I am thrilled,? said Leslie Rosenberg of Hollywood, responding to an inquiry from The Herald. ?Last year, our 22-year-old son was diagnosed with a brain tumor just 10 days after graduating from college. Because of Obama?s health law, we?ve been able to keep him on our employers? insurance policies (despite an extremely high cost) and have him receive outstanding medical care. A year later, he is doing well, and I believe it?s because we were able to have him covered by our policies.?

Al Rothstein, 66, a retired man living in Fort Lauderdale, decried the act as one of the President Barrack Obama moves that ?tend towards socialism,? which will lead to a Republican backlash in November that will kill the act. If that doesn?t happen, the act will lead to ?increased healthcare costs and lowered quality of care,? Rothstein said, responding to an inquiry from The Herald. ?This is a big win,? said Donna Shalala, president of the University of Miami who in the 1990s tried and failed to get healthcare reforms through when she was secretary of Health and Human Services for the Clinton administration.

?It?s a win not just for those who are uninsured but also for those who do have insurance,? because providers have been shifting their costs of treating the uninsured to those who do have coverage. The reform act is ?a first step toward ending cost-shifting. ... It helps every emergency room across the state.?

Healthcare leaders in South Florida were braced for the ruling because the court?s decision is likely to have a huge impact on South Florida, where large numbers of uninsured seeking access to healthcare accounts for $16 billion annually in the region, a larger slice of the region?s revenue than tourism or construction.

When asked how she felt personally, after spending so many years trying to get reforms enacted, Shalala replied: ?It?s thrilling, actually.?

Hospitals, now required to treat the uninsured in emergency rooms even though they rarely get paid for such care, appear to be the biggest winners. Linda Quick, president of the South Florida Hospital and Healthcare Association, sent out a one-word email moments after the decision was announced: ?Yay!?

Think Like a Man Sequel in the Works

Though 2012 has already seen its fair share of box office hits, one of the bigger surprises was the April comedy Think Like a Man.? The film opened at number one at the box office with an impressive take of $33.6 million, ending the four-week run of The Hunger Games and besting new releases The Lucky One and Chimpanzee.? The pic went on to make over $91 million during its run, and now Screen Gems has officially announced that they?re moving forward with a sequel.

No story details are known, but Keith Merryman and David A. Newman will be returning to write the screenplay.? The first film was based on the best selling book by Steve Harvey and featured an ensemble cast dealing with relationships.? No release date has been set either, but expect Think Harder Like a Man to be in front of cameras sooner rather than later.? Jamie Dupree's Washington Insider: Voters knock off another incumbent

While high profile lawmakers like Rep. Charlie Rangel of New York and Sen. Orrin Hatch of Utah survived tough primary challenges, voters on Tuesday night knocked off another member of Congress, as a five term Oklahoma Republican became the latest Congressional casualty.

Rep. John Sullivan (R-OK) wasn't well known outside of his Tulsa-based district, so he wasn't on the radar of many election experts as voting began Tuesday - but by the time the votes were counted, Sullivan was the eighth sitting lawmaker to be dumped by primary voters in 2012.

Compare that to 2010, when only four members lost primary races - or to 2008, when only three were defeated in a primary, and just two in 2006.

Some of the higher numbers in 2012 are directly related to redistricting, which has thrown members into new districts, or even into races involving two sitting lawmakers.

Lawmakers like Jason Altmire (D-PA), Dennis Kucinich (D-OH), Don Manzullo (R-IL) and Steven Rothman (D-NJ) lost those type of primary races to a colleague - and there will be more member vs member races this year.

But others like Sullivan, Rep. Tim Holden (D-PA), Rep. Silvestre Reyes (D-TX) and Rep. Jean Schmidt (R-OH) were also bounced out by the voters for a fresh face - two from each party - indicating a level of discontent that can't be ignored when 19 other states still need to hold their primaries before the November elections.

Sullivan acknowledged recently that he had ignored warning signs from back home about the severity of the challenge; it's a common story of an incumbent who realizes too late that he might be in trouble.

"Biggest lesson of the night for incumbents: Run a campaign," tweeted political analyst Nathan Gonzales, who hit the nail on the head for Sullivan.

We'll see whether the carnage grows the rest of this primary season or not, as members in both parties need to realize, they could end up on the Congressional scrap heap in a hurry.

(AP) ? New York Yankees starter Andy Pettitte broke his left fibula where it meets his ankle and is expected to miss at least six weeks after he was hit by a batted ball during Wednesday's 5-4 win over the Cleveland Indians.

The Yankees say they expect the break to heal without surgery. The injury came on the same day the Yankees placed left-hander CC Sabathia on the 15-day disabled list with a strained groin.

Right-hander Adam Warren is set to be called up from Triple-A on Thursday to make the Friday start that Sabathia was scheduled for. Freddy Garcia is slated to go Monday in Pettitte's spot, and the Yankees are getting David Phelps to pitch deeper into games in the minors with an eye toward bringing him back to the big club as a starter.

General manager Brian Cashman said he didn't want to make a trade to replace Pettitte in the rotation. Garcia has improved his velocity and command recently, and has been an effective starter most of his career. That includes a start in a playoff game he made for New York last season.

"Phelps is a guy, who, if he were stretched out, would be our prime candidate," Cashman said. "We'll just have to figure it out. I would prefer not to go outside."

On the play, Casey Kotchman's low line drive hit Pettitte near his left ankle, the one the left-hander pushes off with. Pettitte took a step toward the ball near the third base line before gently dropping to the infield grass.

Pettitte tried to stay in the game, but manager Joe Girardi removed him when he came up limping after throwing one live pitch. Pettite, who spoke to reporters after the game on crutches ? while clutching a tiny plastic bag with a few white pills in it ? said it hurt too much to push off properly when he was facing a batter.

"I've been hit in the shin, in that area, so many times and I've never had to come out of a game," Pettitte said. "As soon as I threw that first pitch, I had an awful lot of pain, all the way down to my foot."

Pettitte (3-3) retired after the 2010 postseason but returned to the Yankees this spring to shore up New York's pitching rotation, which at the time had already lost Michael Pineda at the time to shoulder surgery.

Pettitte, who has a 243-141 career record, is in his 14th season with New York. He also spent three years with Houston. On both clubs, he was a teammate of Roger Clemens, and he testified at Clemens' recent perjury trial.

With Pettitte out, Derek Jeter is now the only Yankee on the field from the club's Core Four of players who won four World Series titles from 1996-2000. Mariano Rivera is out with a season-ending knee injury and Jorge Posada retired.

"No one's going to feel sorry for us," Girardi said. "Guys have to step up."

A new study shows significant differences in brain development in high-risk infants who develop autism starting as early as age 6 months. The findings published in the American Journal of Psychiatry reveal that this abnormal brain development may be detected before the appearance of autism symptoms in an infant's first year of life. Autism is typically diagnosed around the age of 2 or 3.

The study offers new clues for early diagnosis, which is key, as research suggests that the symptoms of autism - problems with communication, social interaction and behavior - can improve with early intervention. "For the first time, we have an encouraging finding that enables the possibility of developing autism risk biomarkers prior to the appearance of symptoms, and in advance of our current ability to diagnose autism," says co-investigator Dr. Alan Evans at the Montreal Neurological Institute and Hospital the Neuro, McGill University, which is the Data Coordinating Centre for the study.

"Infancy is a time when the brain is being organized and connections are developing rapidly," says Dr. Evans. "Our international research team was able to detect differences in the wiring by six months of age in those children who went on to develop autism. The difference between high-risk infants that developed autism and those that did not was specifically in white matter tract development fibre pathways that connect brain regions." The study followed 92 infants from 6 months to age 2. All were considered at high-risk for autism, as they had older siblings with the developmental disorder. Each infant had a special type of MRI scan, known as diffusion tensor imaging, at 6 months and a behavioral assessment at 24 months. The majority also had additional scans at either or both 12 and 24 months.

At 24 months, 30% of infants in the study were diagnosed with autism. White matter tract development for 12 of the 15 tracts examined differed significantly between the infants that developed autism and those who did not. Researchers evaluated fractional anisotropy (FA), a measure of white matter organization based on the movement of water through tissue. Differences in FA values were greatest at 6 and 24 months. Early in the study, infants who developed autism showed elevated FA values along these tracts, which decreased over time, so that by 24 months autistic infants had lower FA values than infants without autism.

The study characterizes the dynamic age-related brain and behavior changes underlying autism vital for developing tools to aid autistic children and their families. This is the latest finding from the on-going Infant Brain Imaging Study (IBIS), which is funded by the National Institutes of Health (NIH) and brings together the expertise of a network of researchers from institutes across North America. The IBIS study is headquartered at the University of North Carolina, and The Neuro is the Data Coordinating Centre where all IBIS data is centralized.

Biden was responding to a question posed by the Boston Globe, which asked Biden, ?Does executive privilege cover testimony or documents about decision-making within the executive branch not involving confidential advice communicated to the president himself??

Biden delivered an unqualified response.

Biden said that in the same year then-Senator Obama knocked the Bush administration for trying to ?hide behind executive privilege.?

Oil near $79 as Europe woes offset supply threat

(AP) ? Oil fell to near $79 a barrel Monday in Asia as Europe's economic woes offset a supply disruption from a storm that shut down about a quarter of crude output in the Gulf of Mexico.

Benchmark oil for August delivery was down 38 cents at $79.38 a barrel at late afternoon Singapore time in electronic trading on the New York Mercantile Exchange. The contract rose $1.56 to settle at $79.76 in New York on Friday.

In London, Brent crude for August delivery was down 60 cents at $90.38 per barrel on the ICE Futures exchange.

Tropical Storm Debby, with top sustained winds of about 60 mph (95 kph), is lashing Florida and Alabama with heavy rains and is expected to come ashore later this week over the Florida Panhandle. As of Sunday morning, 23 percent of oil and gas production in the Gulf region had been suspended, according to a government hurricane response team. Employees have been evacuated from 13 drilling rigs and 61 production platforms in the Gulf.

Analysts said any supply disruption should be brief since the storm was not expected to significantly damage oil facilities.

Investors will also be closely watching a European Union summit this week where the leaders the leaders of France, Germany, Italy and Spain have agreed to push for a growth package worth up to $163 billion to spur the region's weak economy.

Crude has plunged from $106 less than two months ago amid signs of slowing economic growth and oil demand in the U.S., Europe and China. Capital Economics said it expects Brent crude to fall to as low as $70 during the next 18 months.

"The main downside risk to oil prices comes from the crisis in the euro-zone, which we do expect to worsen," Capital Economics said in a report. "However, after such precipitate declines in oil prices, it would not be surprising to see a small, albeit temporary, bounce."

In other energy trading, heating oil was down 1.1 cents at $2.52 per gallon while gasoline futures gained 0.3 cent at $2.47 per gallon.

Natural gas jumped 5.2 cents at $2.68 per 1,000 cubic feet
